That’s what happens in some cases of Parkinson’s disease, where an enzyme called LRRK2 goes into overdrive. This hyperactivity makes cells lose their “antenna” (primary cilia), blocking dopamine signals and disrupting movement, motivation, and even mood.

Stanford Scientists Hit the Pause Button

The Stanford team wondered if they could cool down this “overheating engine.” They turned to an LRRK2 inhibitor (MLi-2), which works like pressing reset on a jammed button. At first, after two weeks of treatment in mice with the genetic mutation, nothing much happened. But after three months, the story flipped:

neurons regrew their lost “antennas,” and signals began flowing again — like a phone reconnecting to full-bar Wi-Fi after being offline. Even more surprising, these were mature cells, thought to be beyond repair, now showing the ability to self-heal.

The Surprise Comeback

The surprise went beyond just stopping the damage — it hinted at cellular rejuvenation. Results showed:

  • Lower stress signals: dopamine neurons under pressure stopped sending out constant distress calls
  • Neural connections reborn: dopamine nerve endings nearly doubled in density, suggesting stronger brain circuits
  • Protective factors revived: astrocytes and support cells restarted producing neuroprotective proteins, like neighbors rallying together to protect their community

This means the treatment might not just ease symptoms but actively rebuild the brain’s ecosystem.

What This Means for Us

This study suggests Parkinson’s might not only be stabilized but potentially reversed.Early signs often appear 15 years before tremors — like loss of smell, constipation, or acting out dreams in sleep.

With genetic screening and early intervention, this approach could one day support many more patients.
